Depends what kind you mean.
A guy in our mine provides miners.
The Exchange sells them.
You can get some from doff assignments (my trader, Nala Zyal or however you spell her name) keeps getting me green and blue entertainment holograms from blackmailing ferengi. Support colonies in nebulas once you've completed the colony chain will net you a random doff. This is also the best way to pick up colonists.
Buy packs from the c store.
I believe we also have someone stationed at Serenity Station now who will also let you do the 'trade 5 for a single better quality' missions, as well being able to buy fleet special doffs for dil. These two guys are in the room with the Officer of the Watch, bank, mail.
I haven't actually needed to return to Earth to get new doffs in quite some time now. Actually, I haven't been back to earth in ages.

Yes, been able to buy duty officers at Serenity Station for quite some time now. Bit expensive though, I think - not dilithium, actually, fleet credits. Although that's pretty much the same thing, since we ALWAYS need dilithium throwing into some project and that gets you an equal number of fleet credits.
Investigate Intelligence Leak and Raid Safehouse of Facility 4028 Fugitives (possibly others, can't remember) provide a Prisoner on Critical Success. The Prisoner can be quite useful on his own account (especially the Dominion ones), but you can also use the Prisoner Exchange assignment to swap him for a normal good-quality DOff. You can swap Refugees in the same way on the Asylum on So-and-so assigments, but I can't remember where you get Refugees from.

Is there a guaranteed way to get colonists and boot them off your ship? I either have too many or not enough and haven't found a sure fire way to balance it out. The assignments seem completely random but I'm sure there's a method to the madness, somewhere, I hope.

Star clusters and Earth orbit usually have an assignment to pick up colonists (Resettle Colonists) with 100% success. The only mission I've seen that uses them are in the sector clusters, I forget the name but you use 5 of them to set up new colonies. Or they go on the Exchange for a pretty penny.
